Ôř?The Network will expand and/or improve successes achieved by PATC3H to new geographic settings with limited implementation science (IS) research capacity and/or risk populations who are poorly represented in international adolescent HIV research (e.g. sexual and gender minorities; commercial sex workers; drug users) and stimulate much needed IS research in a neglected area of public health significance: prevention of new HIV infections among adolescents at risk and the identification of, and linkage and retention to care of and long-term viral suppression among youth living with HIV in low-to-middle income countries (LMICs). These settings must have an HIV epidemic density defined byÔř?UNAIDS estimatesÔř?as either a country 1) in which at least 200,000 people are living with HIV and the number has not decreased by more than 5% over the last 2 consecutive years of available data or 2) has an HIV incidence among youth ages 10 to 24 years of 0.01% or more.. The structure of PATC3H-IN shall consist of three highly integrated components: (1) 4-5 UG1 Clinical Research Centers (CRC), applications for which have previously been solicited (RFA-HD-23-013); (2) a Coordination and Operations Center (COC); and (3) an Implementation Science Coordinating Center (ISCC).. This Notice serves to announce a forthcoming NOFO that solicits applications for a single Implementation Science Coordinating Center.. HEAL Initiative: Discovery and Validation of Novel Targets for Safe and Effective Pain Treatment (R01 Clinical Trial Not Allowed)

The purpose of this Funding Opportunity Announcement (FOA) is to promote the discovery and validation of novel therapeutic targets to facilitate the development of pain therapeutics. Specifically, the focus of this FOA is on the basic science discovery of targets in the peripheral nervous system, central nervous system, immune system or other tissues in the body that can be used to develop treatments that have minimal side effects and little to no abuse/addiction liability. Research supported by this FOA must include rigorous validation studies to demonstrate the robustness of the target as a pain treatment target. This will lower the risk of adopting the target in translational projects to develop small molecules, biologics, natural substances, or devices that interact with this target for new pain treatments. Translational research to develop new medical devices are not the focus of this FOA. Basic science studies of pain and related systems in the body are responsive to this FOA and are encouraged in the context of novel pain therapeutic target discovery. This FOA is not specific for any one or group of pain conditions. Projects to identify novel targets for acute pain, chronic pain, migraine, other headache disorders, osteoarthritis, diabetic neuropathy, chemotherapy-induced neuropathy, sickle-cell pain, post stroke pain, etc. will be considered. Projects to identify novel targets for a combination of chronic overlapping pain conditions or for specific pathological conditions will be considered. Projects that seek to identify novel targets in specific populations such as women, children, older adults or other underrepresented groups will also be responsive to this FOA.

Global Brain and Nervous System Disorders Research Across the Lifespan (R21 Clinical Trial Optional)

This Funding Opportunity Announcement (FOA) encourages exploratory/developmental research grant applications, proposing the development of innovative, collaborative research projects on brain and other nervous system function and disorders throughout life, relevant to low- and middle-income countries (LMICs). Research on neurological, mental, behavioral, alcohol and substance use disorders may span the full range of science from basic to implementation research. Scientists in the United States (U.S.) or upper-middle income countries (UMICs) are eligible to partner with scientists in LMIC institutions. Scientists in upper middle-income LMICs (UMICs) are also eligible to partner directly with scientists at other LMIC institutions with or without out a US partner. Income categories used are as defined by the World Bank at http://data.worldbank.org/about/country-classifications/country-and-lending-groups. These grants are expected to foster the development of more comprehensive research programs that contribute to the long-term goals of building sustainable research capacity in LMICs to address nervous system development, function and impairment throughout life and to lead to diagnostics, prevention, treatment and implementation strategies. As a starting point, eligible applicants must identify a therapeutic candidate with a robust body of background data in the basic science and early discovery phases to be ready for transition to the preclinical and clinical phases of development. Data may include having sufficient bioactivity, stability, manufacturability, bioavailability, in vivo efficacy and/or target engagement, and other favorable properties that are consistent with the desired clinical application. Projects responsive to this announcement could be undertaken at any point along the drug development continuum, from late discovery (i.e., lead optimization/early safety) up to early-stage clinical trials. For small molecules, the earliest stage of eligibility for this Award is already having small-molecule compounds with proof of desired pharmacological activity. For biologics, the profiling of promising product candidates in animal models of AUD or AAOD will be allowed as the earliest entry point. The ultimate purpose and goal of this FOA is to advance molecules closer to U.S. Food and Drug Administration (FDA) approval.
