Technology Development to Reduce Health Disparities (R01 Clinical Trial Optional)
The purpose of this funding opportunity is to reduce health disparities through the development and translation of appropriate medical technologies. The NIH defines health disparities as differences in the incidence, prevalence, morbidity, mortality, and burden of diseases and other adverse health outcomes that exist among specific population groups. These population groups include racial and ethnic minorities (African Americans, American Indians, Alaska Natives, Asian Americans, Hispanic Americans, Native Hawaiians, and other U.S. Pacific Islanders, as well as subpopulations of all of these racial/ethnic groups), socioeconomically disadvantaged individuals, sexual and gender minorities, and medically underserved populations including individuals residing in rural and urban areas (see https://www.nimhd/nih.gov/about/overview/). This program seeks advances in medical technologies to reduce health disparities associated with diseases, illnesses, and conditions of public health importance. This announcement encourages applications to develop medical devices, imaging systems, robotic systems, biomaterial interfaces, synthetic biological systems, mathematical and modeling solutions, and other technologies to address the healthcare needs of populations that experience health disparities. Proposed medical technologies must have the following basic characteristics: effective, affordable, culturally acceptable, and easily accessible to those who need them. Responsive grant applications will involve a formal collaboration with a healthcare organization or public health agency serving one or more populations that experience health disparities.

Clinical, Behavioral, and Physiological Studies of Open- and Closed-loop Platforms: Toward Personalized, Fully Automated, Accessible Systems (R01 Clinical Trial Required)
The purpose of this fundng opportunity announcemnt (FOA) is to promote clinical research using current and emerging technologies to address barriers that limit progress toward physiological pancreatic hormone replacement open- and closed-loop systems. It will support research to: 1) test and improve the safety, reliability, and clinical efficacy of these technologies; 2) address behavioral/psychosocial factors that play a role in the usability and acceptance of these systems and validation of measures that may be used as outcomes for the demonstration of efficacy and benefit; 3) test these technologies in subpopulations of patients not usually included in clinical trials who may benefit the most from their use; 4) test these technologies in individuals who are underserved or in low income and/or racial and ethnic minority groups, and better understand disparities in the use and adoption of these systems; and 5) use the technologies as tools to advance understanding of glucose regulation and its pathophysiology in patients with type 1 diabetes (T1D) including counter-regulation and impaired awareness of hypoglycemia. Research goals include improved metabolic control with decreased glycemic excursions, prevention of acute and chronic complications, and improved quality of life in patients with diabetes and their caregivers. Only human studies will be considered responsive to this FOA, applications involving animal or in vitro studies are not responsive to this FOA.

In-Depth Phenotyping and Research Using IMPC-Generated Knockout Mouse Strains Exhibiting Embryonic or Perinatal Lethality or Subviability (R01 Clinical Trial Not Allowed)
The purpose of this Funding Opportunity Announcement (FOA) is to encourage applications to phenotype and/or perform research on embryonic lethal knockout (KO) mouse strains being generated through the International Mouse Phenotyping Consortium (IMPC) of which the NIH Knockout Mouse Phenotyping Program (KOMP2) is a member. The mission of IMPC is to generate a comprehensive catalogue of mammalian gene function that will provide the foundation for functional analyses of human genetic variation. The current (July 19, 2022) IMPC data release includes phenotypic data for 8260 knockout genes. Overall, the IMPC hopes to generate a null mutant and undertake broad-based phenotyping for every gene in the mouse genome. About 30% of these strains are expected to be either embryonic or perinatal lethal, or subviable. However, a large portion of homozygous lethal mutations are expected to have viable heterozygous phenotypes. The scientific community has the unique opportunity to leverage these mouse strains while they are being created and bred as part of the IMPC adult mouse phenotyping effort to perform additional in-depth phenotyping and research.

This notice announces the opportunity to apply for funding under the National Organizations of State and Local Officials (NOSLO) Program: State Health Services and Financing to fund a national level organization with an in-depth understanding of, and experience with, providing technical assistance and support to State Medicaid Directors and other health care payment officials.. The purpose of the NOSLO program is to assist state and local authorities in a) improving public health, health care and health care delivery, b) building capacity to address public health issues and support and enforce regulations intended to improve the public‚Äôs health, and c) promoting health equity to preserve and improve public health. To fulfill the purpose, the NOSLO program has the following objectives: 1) facilitate communication and data/information-sharing between HRSA and state, territorial, and/or local officials, 2) improve understanding of state, territorial, and/or local needs, priorities, and perspectives, and 3) support capacity-building activities at the state, territorial, and/or local levels that strengthen the health care safety net and advance public health goals. Pancreatic Cancer Detection Consortium: Management and Data Coordination Unit (U24 Clinical Trial Not Allowed)
The purpose of this Funding Opportunity Announcement (FOA) is to solicit applications for the Research Units (RUs), one of the two scientific components of the Pancreatic Cancer Detection Consortium (PCDC), to conduct research on early detection of pancreatic ductal adenocarcinoma (PDAC) and characterization of its precursor lesions to identify those patients who are at high risk of progression to cancer. The PCDC will continue to address one of the four research priorities identified in the National Cancer Institute's 2014 Scientific Framework for Pancreatic Ductal Adenocarcinoma. The PCDC will support research for the development and testing of new molecular and imaging biomarkers for detecting PDAC early and for identifying those patients at high risk of PDAC (because of genetic factors or presence of precursor lesions) who could be candidates for early intervention. The PCDC-RUs will consist of multi-disciplinary teams and will undertake studies to: identify and test biomarkers measurable in bodily fluids for early detection of PDAC and/or its precursor lesions; determine which pancreatic cysts are likely to progress to cancer; develop molecular- and/or imaging-based approaches for screening populations at high risk of PDAC; use machine learning and computational approaches towards biomarker discovery and/or validation; and conduct biomarker validation studies. The PCDC-RUs will also collect longitudinal biospecimens for building a biorepository. Each PCDC-RU is expected to participate in collaborative activities with other PCDC-RUs and share ideas, biospecimens and data within the Consortium. The other scientific component of the PCDC will be the Management and Data Coordination Unit (MDCU). The PCDC-MDCU will provide support toward study design, protocol development, statistical analysis, coordination, harmonization, data management and stewardship for the trans-PCDC collaborative projects, including biorepository building effort.

Leveraging Social Networks to Promote Widespread Individual Behavior Change (R01 Clinical Trial Optional)
The purpose of this Notice of Funding Opportunity (NOFO) is to invite basic observational or experimental behavioral and/or social science R01 applications that test how intrapersonal and interpersonal mechanisms of behavior change interact with, influence, or are influenced by characteristics of social networks, with implications for health. Research supported through this NOFO will examine at least two levels of analysis: interpersonal processes and social network characteristics. Projects will identify targets for future social network health behavior change interventions across the lifespan, especially in populations in which they are currently largely underdeveloped and untested (such as populations in mid- to- late life). Basic research to develop, refine, or optimize measures (assays) of putative targets (e.g., intra/interpersonal mechanisms of behavior change and/or social network characteristics) are also supported by this NOFO. . Clinical trials that do not meet the federal definition of basic research (CFR 272.3) are not responsive to this RFA. Projects that propose to support the planning activities necessary to develop social network interventions for which a target has already been identified are most appropriate for the R34 RFA companion (RFA-AG-24-026).

This Funding Opportunity Announcement (FOA) encourages applications for competing renewal of Clinical Coordinating Center (CCC) for ongoing clinical trials supported by NCCIH; or competitive revisions to support ancillary studies to ongoing NCCIH supported clinical trials. Extension of an ongoing clinical trial will be supported when there is need for additional time to complete the trial or when an extended period of follow up is well justified to assess longer term outcomes. Competitive revisions will be supported when there is compelling justification for the addition of an ancillary study to an ongoing clinical trial. Rapid advances in genotyping and next generation sequencing technologies have led to the identification of genetic variants that are associated with a wide variety of congenital defects including structural birth defects (SBDs), intellectual developmental disabilities (IDDs) and inborn errors of metabolism (IEMs). Large quantities of genomic data collected from pediatric birth defects cohorts are available to the research community through several databases such as the Database of Genotypes and Phenotypes (dbGaP), the Gabriella Miller Kids First Data Resource Portal, the European Genome-Phenome Archive and Clinical Genome Resource (ClinGen). The National Cancer Institute (NCI) intends to promote a new initiative by publishing a Notice of Funding Opportunity (NOFO) to solicit applications to expand the research scope and leverage the gains made through the Ôř?Investigation of the Transmission of KSHVÔř? initiatives (RFA-CA-18-013 and RFA-CA-20-046)Ôř?to support basic and translational research that will guide the development of a prophylactic or therapeutic Kaposi sarcoma herpesvirus (KSHV) vaccine. A prophylactic KSHV vaccine could prevent primary KSHV infection, transmission, and subsequent development of KS and other KSHV-associated syndromes such multicentric Castleman disease (MCD), primary effusion lymphoma (PEL) and KSHV inflammatory cytokine syndrome (KICS) or ameliorate the severity of disease. A therapeutic KSHV vaccine could be helpful in preventing or treating KSHV disease in people who are already infected with KSHV.. This Notice is being provided to allow potential applicants time to develop responsive projects and meaningful collaborations. The NOFO is expected to use the U01This Notice encourages investigators with expertise and insights into the area of Kaposi sarcoma-associated herpesvirus (KSHV) and/or vaccine development to begin to consider applying for this new NOFO.. Among the areas of research encouraged in this initiative are identification and evaluation of KSHV structural and non-structural targets for a potential KSHV vaccine; development and testing of a candidate KSHV vaccine or vaccines; development of animal models to study a prototype KSHV vaccine or vaccines; studies to assess how the efficacy of a promising KSHV vaccine can be optimized for people with HIV; research to better define the initial steps of infection with KSHV and the primary means of person-to-person transmission in different populations that can be targeted with a vaccine; optimization and/or standardization of KSHV detection methods. In addition, collaborative investigations combining expertise in HIV infection, KSHV infection, immune responses to KSHV and KSHV transmission, virology and vaccine development will be encouraged, and these investigators should also begin considering applying for this NOFO. mechanism with approximately 3-4 awards to be made for each of two receipt dates. Simultaneous and Synergistic Multi-Target Validation for Alzheimers Disease-Related Dementias (R61/R33 Clinical Trial not allowed)
The ultimate goal is to develop multi-target disease-modifying therapeutics based on an appreciation of the complex interaction of pathways, cells, circuits, systems or pathologies that are present in complex AD/ADRD diagnoses. Such therapeutics could be a single drug or biologic targeting multiple pathways, cell, circuits, systems or pathologies, or could be a combination of therapies. While hundreds of possible targets have been suggested for AD alone, theres a need to better understand how multiple targets might interact in a synergistic way that would allow us to tackle AD/ADRD from multiple fronts in a single person, akin to what has been successful in infectious disease or cancer. This FOA will support applications that test hypotheses of possible beneficial interactions of known targets as well as understanding the mechanisms of interaction in the R61. The R33 would support rigorous validation of the proposed multi-target approach (one drug or biologic targeting multiple pathways; and/or a combination of therapies) to support future multi-target therapy development efforts. Successful grantees will be in position to apply for IGNITE or other similar translational grant mechanisms.

This Forecast is modified to correct the Estimated Total Funding from $9,000,000 to $4,719,633. The Estimated Post Date and Estimated Application Due Date have remained the same.The purpose of the Competitive SRAE Program is to fund projects to implement sexual risk avoidance education that teach participants how to voluntarily refrain from non-marital sexual activity. Successful applicants are expected to submit plans for the implementation of sexual risk avoidance education that normalizes the optimal health behavior of avoiding non-marital sexual activity, with a focus on the future health, psychological wellbeing, and economic success of youth.  Applicants must agree to: 1) use medically accurate information referenced to peer-reviewed publications by educational, scientific, governmental, or health organizations; implement an evidence-based approach integrating research findings with practical implementation that aligns with the needs and desired outcomes for the intended audience; and 2) teach the benefits associated with self-regulation, success sequencing for poverty prevention, healthy relationships, goal setting, resisting sexual coercion, dating violence, and other youth risk behaviors such as underage drinking or illicit drug use without normalizing teen sexual activity. The Title V SRAE legislation requires unambiguous and primary emphasis and context for each of the A-F topics listed in the NOFO to be addressed in program implementation. Additionally, there is a requirement that messages to youth normalize the optimal health behavior of avoiding non-marital sexual activity.
